Effect of steroid nandrolone decanoate on osteoblast-like cells

Background: Bone is a target tissue of the action of sex steroids, but the mechanisms by which androgens exert their effects on bone biology are not yet completely understood. The objective of the present study was to evaluate the effects of the anabolic steroid nandrolone decanoate on the viability, adhesion, and alkaline phosphatase activity of osteoblasts-like cells.
Material and Methods: Cell viability and cell adhesion were evaluated using the MTT method. Alkaline phosphatase activity was measured in lysed cells using a commercial kit.
Results: The results revealed a significant decrease in the viability of osteoblasts treated with 50 µM of nandrolone decanoate after 72 h of culture; however, alkaline phosphatase activity increased significantly in this group in comparison with the control group. Moreover, a significant increase in cell adhesion was found in the groups treated with 25 µM of nandrolone decanoate after 40 and 60 min of incubation.
Conclusions: Besides exerting an anabolic effect on muscle tissue, treatment with nandrolone decanoate may promote effects on bone by increasing adherence and alkaline phosphatase activity of osteoblasts-like cells.
